V prispevku je opisana metoda kovalentne imobilizacije tripsina
na povrsino asimetricne porozne membrane iz celuloznega acetata.
Dolocene so pretocne lastnosti in debeline membrane pred
imobilizacijo encima in po njej, aktivnost tripsina v raztopini
in lastnosti tripsina v obmocju pH od 2 do 12. Potek hidrolize
povrsine membrane smo potrdili s primerjavo ATR spektrov pred
hidrolizo in po njej. Aktivnost imobiliziranega tripsina smo
dolocili z Anson-ovo metodo in jo primerjali z aktivnostjo
tripsina v raztopini.

In this paper the method for covalent immobilization of Trypsin
on the surface of asymmetric porous membrane from cellulosic
acetate is described. Flux properties and thickness of the
membranes before and after immobilization have been determined.
Activity of the Trypsin in the solution and the properties of the
Trypsin in pH range from 2 to 12 have been estimated. Hydrolysis
of the surface of membrane has been confirmed by comparison the
ATR spectrum before and after hydrolysis. The activity of
immobilized Trypsin has been determined by using Anson procedure
and it has been compared with the activity of Trypsin in the
solution.
